Essay

The aim of this work was to investigate glucosaminyl-muramyl dipeptide (GMDP) both free and liposomal form pharmacokinetics and biodistribution, liposomal GMDP influence on activity of macrophages and antimetastatic effects. For the first time liposomal peptide was demonstrated to modulate macrophage nitric oxide synthesis activity in dose dependent mode by nitrite and nitrate definition as final metabolites. Nitrite and nitrate production by macrophages in response to LPS was found to be enhanced by liposomal GMDP at lower concentrations (0.02 mkg/ml) and suppressed at higher (up to 0.5 mkg/ml). The specific features of pharmacokinetics and modes of tissue macrophages activity modulation are anticipated as key requirement for effective liposomal GMDP use in anticancer immunotherapy.
